Knowing of bacteriophage in laboratory     

For the growth of bacteriophage, we can use either a liquid or solid medium.

Procedure 

1. Mix melted agar with bacteriophage (T4) suspension and host bacteria (E-coli).
2. Take Petri plate and pour MacConkey agar on it.
3. Pour mixture on agar.
4. Incubate the plate, after a certain incubation period, there is the formation of the clear zone on the plate.
5. Cut agar with a sharp blade and place it in chloroform in a test tube. It will separate cell debris and the media part from bacteriophage.
6. Collect virus by filtration.
